Eli Lilly announced topline results from TRIUMPH-1, the first pivotal Phase 3 trial of retatrutide in adults with obesity or overweight without diabetes. The results exceeded expectations — and the bar set by existing treatments.
The numbers
Retatrutide achieved mean weight reduction of up to 30.3% from baseline in the highest dose group, meeting the trial’s primary and key secondary endpoints with statistical significance. For context, currently approved treatments achieve roughly 15–21% mean weight loss in their registration trials.
At the highest dose:
- Over 90% of participants achieved at least 5% weight loss
- A majority achieved reductions exceeding 20%
- The weight loss trajectory had not plateaued at the end of the treatment period
Safety
The safety and tolerability profile was consistent with the known incretin class and with retatrutide’s Phase 2 data. Gastrointestinal side effects — nausea, diarrhea, vomiting — were the most common, generally mild to moderate, and improved over time. Context
TRIUMPH-1 is the first of five Phase 3 trials in the TRIUMPH program to report results. It enrolled adults with obesity or overweight with at least one weight-related comorbidity, excluding type 2 diabetes. (A separate trial, TRIUMPH-2, covers the type 2 diabetes population.)
These results are the strongest weight loss data reported for any pharmaceutical obesity treatment in a Phase 3 trial. The 30.3% figure approaches what is typically seen with bariatric surgery, a comparison that generated substantial media coverage and medical community discussion.
